Resumo Vários fatores podem levar a alterações da qualidade da água em ambientes aquáticos, levando a inúmeras consequências para os organismos que os habitam. Os corpos d'água localizados na região noroeste do Rio Grande do Sul recebem periodicamente poluentes que podem conter propriedades genotóxicas, citotóxicas e mutagênicas, que afetam a integridade genética dos organismos que ali vivem. Esta pesquisa teve como objetivo analisar as respostas citológicas de Danio rerio exposto à água doce antropizada do rio Ijuí pelo método do micronúcleo, bem como pela observação de anormalidades nucleares em seus eritrócitos. Os dados coletados demonstraram valores maiores de eritrócitos com NA do que com MN, sugerindo que as células são eficientes nos mecanismos de reparo necessários ao enfrentamento de substâncias genotóxicas.

Abstract Several factors can lead to alterations of water quality in aquatic environments, leading to numerous consequences for the organisms that inhabit it. The water bodies located in the northwest region of Rio Grande do Sul periodically receive pollutants that may have genotoxic, cytotoxic and mutagenic properties, which affect the genetic integrity of organisms living there. This research analyzed the cytological responses of Danio rerio exposed to anthropized fresh water from the Ijuí River by the micronucleus method as well as by observing nuclear abnormalities in their erythrocytes. The data collected demonstrated higher values of erythrocytes with NA than with MN, suggesting that the cells are efficient in the repair mechanisms required when facing genotoxic substances.

Resumo Inúmeros fatores de poluição geram alterações na qualidade da água e nos organismos que dela dependem, visto que podem conter substâncias com propriedades mutagênicas, genotóxicas e citotóxicas que afetam a integridade genética dos organismos. No município de Cerro Largo - RS localiza-se o Arroio Clarimundo, que no seu fluxo passa por uma área agrícola e atravessa a cidade recebendo descarga de resíduos domésticos não tratados. Este trabalho teve como objetivo, avaliar o potencial genotóxico dessas águas por meio do bioensaio com Allium cepa usualmente empregado em estudos de biomonitoramento. Para a realização das análises foram feitas quatro coletas de água, em cada estação do ano e em três pontos demarcados. Os bulbos de Allium cepa foram postos para germinar, durante 72 horas, na proporção de três para cada amostra dos pontos de coleta e três em água destilada para parâmetro de controle. Posteriormente, as raízes foram submetidas ao protocolo de Feulgen com modificações, para a confecção das lâminas. Foram observadas alterações nos valores de Índice Mitótico e Alterações Cromossômicas. Os dados obtidos foram submetidos a análise estatística pelo teste de Tukey com confiabilidade de 5%. Os resultados sugerem que a água do Arroio Clarimundo tem potencial para causar danos no genoma de organismos eucariotos.

Abstract Numerous pollution factors generate changes in water quality and in organisms that depend on this resource, as they may contain substances with mutagenic, genotoxic and cytotoxic properties that affect the genetic integrity of organisms. The Clarimundo Stream is located in the city of Cerro Largo, Rio Grande do Sul, Brazil, from which it passes through an agricultural area and crosses the city receiving discharges of untreated household waste. This work evaluated the genotoxic potential of these waters through a bioassay with Allium cepa, usually used in biomonitoring studies. Four water collections were conducted in each season and three points were marked for the analyses. The Allium cepa bulbs were germinated for 72 hours, in the proportion of three for each sample of collection points and three in distilled water for control parameters. Subsequently, the roots were submitted to the Feulgen procedure with modifications, for the preparation of the slides. The data were submitted to statistical analysis by the Tukey test with reliability of 5%. The results showed chromosomal alterations and Mitotic Index values suggesting that the water of Clarimundo Stream has the potential to cause damage to the genome of eukaryotic organisms.

As amilases estão entre as mais importantes enzimas industriais, apresentando grande importância biotecnológica, principalmente na indústria alimentícia. Com o avanço no conhecimento das enzimas, a utilização dos fungos como fonte de enzimas vem adquirindo um status de destaque nas mais variadas áreas industriais e comerciais. Diante disso, o presente estudo procurou identificar a presença de atividade amilolítica em quatro linhagens do fungo filamentoso Aspergillus nidulans, selvagem, PAT, biA1methG1 e CLB3, utilizando dois meios distintos de cultura, BDA e Meio Completo a 2% amido, variando os tratamentos com adição ou não de glicose. Foram determinados o diâmetro médio da colônia, o diâmetro médio do halo e o Índice Enzimático. Como resultados, todas as linhagens testadas foram capazes de degradar o amido quando na ausência de glicose, porém o tratamento que obteve estatisticamente melhor crescimento e maior degradação do amido foi o MC sem glicose a 2% amido e a linhagem que se demonstrou potencialmente degradadora de amido foi o mutante CLB3. Conclui-se, portanto, que Aspergillus nidulans pode ser considerado como um produtor de amilases.

The amylases are among the most important industrial enzymes showing great biotechnological importance, mainly in the food industry. Due to the advance knowledge in enzyme research, the use of fungi as source of enzymes has acquired a prominent status in most varied industrial and commercial areas. Thus, the present study tried to identify the presence of the amylolytic activity in four stains of filamentous fungi Aspergillus nidulans, selvagem, PAT, biA1methG1 and CLB3 by using two different culture means, PDA and Complete mean with 2% starch, varying the treatments with and without the addition of glucose. The colony average diameter, the halo average diameter, and the enzymatic index were determined. The results show that all strains tested were capable of degrading starch in the absence of glucose although the treatment that presented statistically better growing and bigger degradation of starch was CM without glucose with 2% starch and the strain that demonstrated potentially degradation was CLB3 mutant. It was concluded that Aspergillus nidulans can be considered as an enzyme producer.
